For example, consider Turks who live in Turkey on one hand and consider ethnic Turks who have been living in Germany. Assume that, all the German national Turks always marry each other, so do the Turks in Turkey. After so many generations, would there be a significant genetic difference between those two groups?

Turks haven't been in Germany nearly long enough for this to happen, and hopefully they will be kicked out of Germany soon and return to Turkey.

i think it can happen what happened for italian americans.

since the most of italian americans were Southern italians with a big bunch of Sicilians in the end italian american commmunity got darker pigmentation in comparison with Italy and even slighly darker than average modern south italian

Yes it does happen but it takes a lot of isolation and drift. It happened with Irish Travellers and the settled population. Irish Travellers have their own cluster away from the main Irish population. Irish Travellers are thought to have taken to the road around the time of the British plantation of Ulster in the 17th Century. They have practiced endogamy and have largely kept to themselves. They are a small population though only about 50,000 approx and I think the Turkish population in Germany is massively huge compared the Travellers. You can also see that Orkney has drifted from the main British Isles population cluster.
